World Parkinson's Disease Day: Role of Genetics and Environment in Parkinson's Disease
Parkinson's disease is a neurodegenerative disorder, with age being the most significant risk factor. While genetics play a role in Parkinson's disease, environmental factors, such as exposure to toxins like pesticides, herbicides, heavy metals like lead and mercury, head injuries, viral infections, and inflammation, may also cause it. Specific genetic mutations have been identified in rare cases of Parkinson's disease, but the sporadic type without family history results from a convoluted relation of genetic and environmental factors. According to Dr Guruprasad H, Senior Consultant Neurologist, Manipal Hospital, the onset of Parkinson's disease is mostly in the younger age group for familial PD, where substantial causative genetic mutations have been identified. The loss of dopaminergic cells in the midbrain with the growth of Lewy body inclusions in brainstem cells is the hallmark of Parkinson's disease pathogenesis. Mutation of genes leads to mitochondrial dysfunction, oxidative stress, accumulation of misfolded proteins, slowness, immobility, tremors and programmed cell death of affected neurons and non-motor symptoms, such as depression, constipation, sleep issues and more.
